Leibovitz's L-15 Medium was originally designed to be used in carbon dioxide (CO
2) free systems and therefore does not need sodium bicarbonate to provide the buffering capacity. Instead, phosphates, free base amino acids, salts, and galactose maintain the physiological pH.
Application of Leibovitz's L-15:
- Cryopreservation of cells and tissues such as Xenopus sperm and embryonic cells.
- Establishing human tumor cells lines such as Hep-2 cells.
- As a supplement in sterile cell media for the dissection and isolation of kidney cells.
- As an important media which provides proper buffering system for culturing of lung cells.
- A valid option for growing fish originated cell lines
This media has unique criteria:
- High Level of Sodium pyruvate (5mM)
- Absence of Sodium Bicarbonate which is commonly used in other media for maintaining buffering capacity.
- Using Galactose instead of glucose
Leibovitz's L-15 contains no proteins or growth factors and therefore should be supplemented with FBS.
