PurMaTM  10X Green Dye is a Loading Dye formulated to prepare and to facilitate loading of DNA or RNA containing solutions of marker and samples in the wells of agarose and non-denaturing polyacrylamide gels for electrophoresis. The loading dyes add color and increase the density of the samples, thus simplify the loading process. Green dye is a combination of Yellow and blue dye that made from Orange G and Xylene Cyanol FF (at a low concentration) respectively that separate and yield two clear blue and yellow bands as the dyes move toward the anode at distinct rates in the electric field during electrophoresis.

Description

PurMaTM  10X Green Dye is a Loading Dye formulated to prepare and to facilitate loading of DNA or RNA containing solutions of marker and samples in the wells of agarose and non-denaturing polyacrylamide gels for electrophoresis. The loading dyes add color and increase the density of the samples, thus simplify the loading process. Green dye is a combination of Yellow and blue dye that made from Orange G and Xylene Cyanol FF (at a low concentration) respectively that separate and yield two clear blue and yellow bands as the dyes move toward the anode at distinct rates in the electric field during electrophoresis. These two different tracking dyes act as indicator for DNA fragment migration on agarose and polyacrylamide gels and help in visual tracking to monitor the progress of migration during electrophoresis. Orange G dye (the yellow dye) migrates at a faster rate with the smallest DNA fragments (<50bp) whereas xylene cyanol FF (the blue dye)  migrates much slower than yellow at approximately similar rate of 3000-5000 bp DNA fragments on a standard 1% TAE or TBE agarose gels. (or 800 bp and 30 bp on a 2% TAE agarose gel respectively). Xylene Cyanol FF, the blue dye is useful in tracking comparatively large DNA fragments for an extended period of time even long after orange G has run off the bottom of the gel. While the yellow dye helps in monitoring small DNA fragments. As yellow dye migrates quickly with the level of <50bp DNA fragments, we can be ensured the DNA fragments that are larger than 50bp is safely retained within the gel as long as the yellow dye is still visible.

The dyes are non-interfering and PCR performance is not negatively influenced with the dye at all. During electrophoresis the tracking dyes migrate without masking the DNA fragments under UV light within the samples.

PurMaTM 10X Green Dye is used in a wide variety of PCR applications including RT-PCR or q PCR. It can also be used in PCR based downstream applications like DNA sequencing, ligation and restriction digestion. But it is not recommended for the PCR applications that use any absorbance or fluorescence excitation, as the blue and yellow tracking dyes included in the mixture may interfere with those process.

In addition to the tracking dyes, PurMaTM 10X Green Dye contains 60% glycerol and EDTA. The presence of glycerol increases the density and ensures that the DNA in the samples and markers forms a layer at the bottom of the well nicely.  EDTA also included in the solution that chelates divalent metal ions to protect DNA from metal dependent nucleases that might be activated from metal dependent enzymatic reactions.

 

PurMaTM 10 X Green Dye contains:

— 0.03% Xylene cyanol FF

— 0.15% Orange G

— 60 % (w/v) Glycerol,

–60 mM EDTA,

Usage notes

 

Add 1:10 loading Dye with DNA sample as shown in the table below. Mix properly to achieve homogeneity.
