EMEM (Eagle’s Minimal Essential Medium) is one of the most widely used basal media in cell culture. Developed by Harry Eagle as a refinement of his earlier Basal Medium Eagle, it supplies the salts, amino acids, vitamins, and glucose that a broad range of mammalian cells need to grow, once it is completed with serum and glutamine. The word “minimal” points to its lean, well-defined formulation: it provides the essential nutrients cells require, without the extra components found in richer media like DMEM.

What is EMEM?
EMEM is a chemically balanced basal medium designed to support the growth of adherent mammalian cells. It starts from a balanced salt solution and adds the essential amino acids and vitamins that cells cannot make for themselves in sufficient amounts. Because it is a basal medium, EMEM is not complete on its own; researchers add serum (commonly FBS), L-glutamine, and often non-essential amino acids before use.
What is in EMEM?
A typical EMEM formulation contains:
- A balanced salt solution that maintains osmotic pressure and pH (Earle’s or Hanks’ salts, more on this below)
- Essential amino acids, the building blocks cells cannot synthesize in sufficient quantity
- Vitamins (the Eagle’s vitamin set) that serve as metabolic cofactors
- Glucose as the primary energy source
- A bicarbonate buffer and usually phenol red as a pH indicator
EMEM was formulated with higher amino acid concentrations than earlier media to better match the composition of mammalian cells, which improved growth and reproducibility.
Earle’s vs. Hanks’ salts: which EMEM do you need?
EMEM comes with one of two balanced salt solutions, and the choice matters:
- EMEM with Earle’s salts uses a higher bicarbonate concentration and is buffered by a CO2 atmosphere. Use it in a CO2 incubator (typically 5 to 10% CO2).
- EMEM with Hanks’ salts uses a lower bicarbonate level and is intended for closed systems without added CO2.

What is EMEM used for?
EMEM is a workhorse basal medium used across cell biology, including:
- Growing adherent mammalian cell lines such as fibroblasts and many epithelial lines
- Primary cell culture, where a leaner, well-defined medium is often preferred
- Virology and vaccine work, a long-standing application for MEM-type media
- Routine maintenance of cells that do not require a richer formulation
EMEM vs. DMEM
EMEM and DMEM are closely related, but DMEM (Dulbecco’s Modified Eagle’s Medium) is a richer modification: it contains roughly four times the amino acids and vitamins of EMEM and is offered in higher-glucose formulations. As a rule of thumb, DMEM suits dense, fast-growing, or demanding cultures, while EMEM’s leaner formulation fits cells that grow well without the extra nutrients. How to supplement and use EMEM
Before use, EMEM is typically completed with:
- Serum (commonly 5 to 10% FBS) for growth factors and attachment
- L-glutamine or a stable glutamine dipeptide, since glutamine degrades over time
- Non-essential amino acids, and sometimes sodium pyruvate, depending on the cell line
Store EMEM refrigerated and protected from light, warm it gently before use, and keep the same lot across an experiment for consistency. If you use Earle’s-salt EMEM, remember it needs a CO2 environment to hold the correct pH.
