- Serums are the source of early stages of some pathogens such as micrococcus. The small structure of these elements causes them passing through 0.22 µm. They adhere to flask and dishes and after about one week, they turn into mycoplasma.
- The color of the media has NOT changed, and the media won’t be turbid. Whereas, with bacterial, mold, fungus contamination, you could clearly see the contamination.

In cell culture the acceptable level of endotoxin is 0.5 units/ml which only achieved by complete attention and awareness of the nature and causes of endotoxin contamination. As sterile conditions are not always provided, endotoxin contamination during in vitro work is very common. The most common source of endotoxin is water since distillation and deionizing columns do not remove endotoxin. In in vitro studies, endotoxins may be contained in buffers and/or on materials and affect observed results. the presence of relatively high levels of environmental endotoxins (100 EU/ml) has a dramatic effect on the results. That is because they generate wear debris in vitro and have confirmed the significant effect of environmental endotoxin contamination on cytokines which is one of the main players of intracellular signaling in cell culture process and directly affect the nuclear phosphorylation of CREB.
There is various type of endotoxins ranging from molecular weights of 10,000 to 1,000,000. However, since proteins are in the same molecular weight range as endotoxins, usual sterilization such as filtration cannot eliminate the endotoxins from the mixture of proteins. That brings us to the conclusion that preventing endotoxin contamination by purchasing chemicals, cell culture reagent and water from a reputable vendor is still the best strategy to fight endotoxins. As endotoxins cause the proteins to have negative charge in most solutions, ion exchange chromatography is an option to decontaminate protein from these contaminants.
It thoroughly depends on the cell lines. The heat inactivation of FBS is unnecessary for most of the cell lines. In has been proven to us that the differentiated cell line do much better in heat inactivated serum.
The best way to find out whether heat inactivation is necessary for your cell culture is to empirically determine that in your own study.
