- Basically, the entire procedure with PurMaFectin™ Ultra is completed within 30 minutes.
- Furthermore, for the highest efficacy and the lowest dead cells, transfect cells at around 70~80% confluency. We strongly recommend starting the seeding by counting the trypsinized cells so there will be high consistency among your experiments.
- Additionally, different cell types react differently to the number of passages when they are used for transfection. So, to minimize the impact of passage on efficiency of transfection, use the cells with the same passage number, or alternatively, apply at least two different concentrations of transfection reagent as control in new transfect experiments before the condition is optimized.
- We deplete endotoxin from our transfection reagent. The endotoxin-contaminated DNA results in inefficient transfection and can cause high cellular toxicity. To deplete the endotoxin, we use PurMaTM Endotoxin Elimination Column (Cat# P6H1216251). In this proprietary kit, Sepharose 6B covalently conjugates to an endotoxin-specific absorbent. The mentioned procedure will end with passing the reagent through 0.1µm filters 3X (included in the kit). The mentioned procedure eliminates the residual endotoxin.

Frequently asked questions
Should serum be present during transfection?
It depends on the reagent. Many lipid-based reagents form complexes better in serum-free medium, with serum added back after a few hours. Check the protocol for the specific product rather than assuming.
How do I optimise the reagent to DNA ratio?
Run a small matrix on your specific cell line, varying reagent volume against a fixed DNA amount. The optimum differs markedly between lines and is the single biggest determinant of efficiency.
Why is my transfection efficiency low?
The usual causes are cell confluence outside the optimal window, poor quality or endotoxin-contaminated DNA, too little or too much reagent, and antibiotics in the medium during transfection increasing toxicity.
Is the reagent toxic to cells?
All transfection reagents carry some toxicity, which rises with concentration and exposure time. If viability drops sharply, reduce the reagent amount and shorten the exposure before assuming the cells are the problem.
How should PurMaFectin™ Ultra Transfection Reagent be stored?
At the temperature stated on the Certificate of Analysis, usually 2 to 8 degrees C, and not frozen unless the label says so. Freezing many lipid formulations destroys them irreversibly.
