What is cryoprotective media?

Cryoprotective media is a specially formulated solution that protects cells from the damage caused by freezing and thawing. During freezing, ice crystals can form inside and around cells and rupture them, and the concentration of solutes rises sharply. Cryoprotective media contains agents that limit this damage, so a high fraction of cells stay viable after storage in a freezer or liquid nitrogen.

How DMSO protects cells

DMSO (dimethyl sulfoxide) is the most widely used cryoprotectant. It penetrates cells and reduces ice crystal formation, lowering the mechanical and osmotic stress of freezing. A typical formulation uses around 5 to 10% DMSO, often with serum or a serum substitute. Because DMSO can be toxic at room temperature, cells should be frozen promptly after it is added and diluted out quickly after thawing.

How to freeze and thaw cells

For freezing, suspend healthy cells in cryoprotective media, transfer to cryovials, and cool them slowly (about 1 degree C per minute, often using a controlled-rate container) before moving them to long-term storage. For thawing, warm the vial quickly in a 37 degree C bath, then gently dilute the cells into warm complete media to remove the cryoprotectant. Handle cells gently right after thawing, since they are fragile.
