DMEM-F-12
(Dulbecco’s Modified Eagle Medium Nutrient-Mixture F-12) provides superior quality, consistency, and great control in mammalian cell culture.
This formulation combines DMEM’s high concentrations of glucose, amino acids, and vitamins as well as much more variation of salts with DMEM-F-12 ’s wide variety of components.
This media is better than DMEM and advanced IMDM on MDCK, glial cells, fibroblasts, human endothelial cells, and rat fibroblasts. It contains no proteins, lipids, or growth factors. Therefore, it requires supplementation, commonly with 10% Fetal Bovine Serum (FBS).
In general, DMEM- F-12 is more enriched but contains the following major changes compared to DMEM:
- 2.5 mM L-glutamine,
- 0.5 mM sodium pyruvate,
- 30mM NaHCO3 (2.438g/L)

- Phenol Red
- L-glutamine
In some formulations, the concentration of sodium bicarbonate used is lower than DMEM, therefore requires a 5–10% CO2 environment to maintain physiological pH.
