Background

During cryopreservation cooling aqueous solutions below the equilibrium freezing point will inevitably result in the formation of ice crystals in the extracellular media. PurMa™ Serum Free Cryoprotective Media With DMSO allows the cryoprotective agent to get inside the cells instantly and lower cell death because of penetration / freezing in absence of any serum or animal product. That will inevitably,  cause a decrease in concentration of water which has two key consequences:

  1. Osmotic gradient forms across the cell membrane, which causes “cell dehydration”, and
  2. Ice formation and abnormal concentration of solutes in intracellular matrix, osmotic shock, and toxicity.

Serum in Cryoprotective Media

Serum and its components perform a crucial role in cells viability as well as recovery during cryopreservation. Nevertheless, there is known disadvantages of the serum in Cryoprotective media:

  1. The inability to fully sterilize them.
  2. Moreover, unknown composition.
  3. And finally, the batch-to-batch variation which results in inconsistency in the rate of survival, recovery, and viability in cryopreservation. Additionally, more crucial for sensitive cells to cryopreservation.
  4. Therefore, a chemically defined Cryoprotective media resolves the several including the above-mentioned restrictions.

Two Major Benefit

  1. PurMaTM Serum Free Cryoprotective Medium (SFCM) prevents cell hydration and formation of ice crystals, therefore is an excellent tool for short- and long-term cryopreservation.
  2. PurMaTM Serum Free Cryoprotective Medium (SFCM) contains DMSO.

DMSO benefits cryoprotection and cryoprotective agent in several ways

  1. Provides strong molecular stability.

  3. It contains the basic properties, salts solubility particularly for anions. Additionally contains a propensity to act as the acceptor atom in hydrogen bonding.
  4. Especially, DMSO makes a  hydrogen bond with water which is 30% stronger than between two water molecules.
