Background

During cryopreservation cooling aqueous solutions below the equilibrium freezing point will inevitably result in the formation of ice crystals in the extracellular media. PurMa™ Serum Free Cryoprotective Media With DMSO allows the cryoprotective agent to get inside the cells instantly and lower cell death because of penetration / freezing in the absence of any serum or animal product. That will inevitably cause a decrease in the concentration of water, which has two key consequences:

  1. Osmotic gradient forms across the cell membrane, which causes “cell dehydration”, and
  2. Ice formation and abnormal concentration of solutes in intracellular matrix, osmotic shock, and toxicity.

Serum in Cryoprotective Media

Serum and its components perform a crucial role in cells viability as well as recovery during cryopreservation. Nevertheless, there are known disadvantages of serum in cryoprotective media:

  1. The inability to fully sterilize them.
  2. Moreover, unknown composition.
  3. And finally, the batch-to-batch variation which results in inconsistency in the rate of survival, recovery, and viability in cryopreservation. Additionally, this is more crucial for cells sensitive to cryopreservation.
  4. Therefore, a chemically defined cryoprotective media resolves several issues, including the above-mentioned restrictions.

DMSO benefits cryoprotection and cryoprotective agent in several ways

Should I choose the DMSO or DMSO free formulation?

DMSO is the standard cryoprotectant and gives the best recovery for most lines. Choose DMSO free where DMSO would interfere with the application, for example in differentiation work or certain clinical and hybridoma protocols where residual DMSO is a problem.

What freezing rate should I use?

Approximately minus 1 degree C per minute to around minus 80, using a controlled rate cooler or an insulated freezing container, before transfer to liquid nitrogen. Faster cooling causes intracellular ice and slower cooling causes osmotic damage.

How should vials be stored long term?

In the vapour phase of liquid nitrogen, below minus 150 degrees C. Storage in a minus 80 freezer is acceptable only for weeks, not years, because viability declines steadily above minus 130.

How should I thaw cells frozen in Serum Free Cryoprotective Media With DMSO?

Thaw rapidly in a 37 degree C bath until only a small ice crystal remains, then dilute promptly into pre warmed medium. DMSO is toxic to cells at room temperature, so the interval between thaw and dilution should be as short as possible.

How should the cryoprotective media itself be stored?

Store as stated on the Certificate of Analysis, protected from light. Do not refreeze after thawing the media, and use within the period stated for the lot.
