Description
Background
PurMa™ Trypsin-EDTA 5% contains ultra-pure trypsin. Furthermore, trypsin is one of the members of the serine protease family which goes through a set of intracellular signaling that results in the activation of this enzyme in the pancreas. Cell culture laboratories widely use trypsin to dissociate the adherent cells for routine cell culture. The Trypsin family is conservative. Trypsin performs the best at 37°C and at the PH 7.3-8.5.
Trypsin Activity
Trypsin cleaves its substrates: IGDKLIPA unless these amino acids are followed by proline. PurMa™ Trypsin is HPLC purified 99% purity for bovine pancreas. Moreover, optical rotatory dispersion of trypsin in the presence of calcium shows a major conformational transition between 40 and 45° which results in deactivation of trypsin. Therefore, all the buffers needed to be Ca+2 & Mg+2 free for the proper function of this enzyme.
PurMa™ Trypsin Deactivates Mycoplasma in your Cell Culture
Trypsin treatment abolishes the ability of mycoplasmas to attach to the cells. Mycoplasma is one of the common and slowly deteriorating contaminations in cell culture, especially due to the invisibility of the contamination. PurMa Biologics manufactures all the reagents you need to fight mycoplasma in your cell culture. The presence of a trace of SDS and other detergents leads to a decrease in the trypsin activity.
Preparation
The activity of this enzyme is further modulated by the sulfation of Tyr154. What sizes does Trypsin-EDTA 5% come in?
Trypsin-EDTA 5% is supplied in 1 x 100 mL, 6 x 100 mL, 1 x 500 mL and 6 x 500 mL, with or without phenol red.
Is 5% trypsin the right concentration for routine passaging?
For routine passaging of most adherent lines, 0.05% or 0.25% Trypsin-EDTA is the usual choice. 5% is a concentrated formulation intended for dilution or for particularly stubborn tissue and primary preparations. If you are passaging a standard cell line, the 0.05% or 0.25% products are likely the better fit.
Can I get it without calcium chloride or sodium bicarbonate?
Yes. Alongside the standard formulation, Trypsin 5% is available without sodium bicarbonate, without calcium chloride and without glucose.
Is it sterile, and do you provide a Certificate of Analysis?
Yes. Every lot is manufactured and quality controlled in house in Oklahoma City, and a Certificate of Analysis is available for every lot. Storage conditions and the expiry date for your specific lot are printed on the bottle and stated on that lot's COA.
Can you supply trypsin in bulk or on a purchase order?
