Standard Dulbecco’s Modified Eagle’s Medium (DMEM) uses L‑glutamine supplementation. DMEM‑MAX replaces L‑glutamine with 4 mM L‑alanyl‑L‑glutamine, which greatly enhances stability and reduces toxic by‑products.

Why L‑Alanyl‑L‑Glutamine Instead of L‑Glutamine? (Mechanism)

While L‑glutamine is an essential amino acid for mammalian cells, it is chemically unstable in aqueous solution. Over time, L‑glutamine degrades and generates toxic ammonia and pyrrolidone carboxylic acid, which can:

  • Stress or damage cells which is proven to be far less in Dulbecco’s Modified Eagle’s Medium Max
  • Reduce viability and growth
  • Compromise experimental reproducibility

One way to address this is to frequently add fresh L‑glutamine to the medium, but this is labor‑intensive and difficult to control precisely.
