Background

Since the serum usually  presents in cell culture studies,  the lipid metabolism makes studying the role of serum in cultured cells very challenging.  That is because serum contains a wide range of undefined lipid species. PurMa™ Lipid-Depleted Fetal Horse Serum (LD-FHS) performs as the highest level of lipid depletion of serum . Lipid-Depleted Fetal Horse Serum (LD-FHS) performs crucial roles in:

  1. Where the normal concentration of lipids may interfere with the results.
  2.  It Pays  also crucial for studies directly involving lipid metabolism. Because the composition of serum can have a profound impact on important cellular checkpoints.
  3. Moreover, LD-FHS performs in investigations about the role of lipids in their corresponding intracellular pathways.
  4. Also, it acts to generate  the media with defined lipid species.

PurMa Biologics uses a cutting-edge methodology to remove lipids from serum without eliminating its ability to support the proliferation of cells in culture.

Our proprietary method includes

  1. Applying isopropyl ether and butanol with a special ratio.
  2. PurMa Biologics has spent many years determining the needed density of ether and butanol as well as the ratio of these two solvents in this process to gain the best result.
  3. 4X washed each 3h hours with isopropyl ether-butanol with decreasing amount of butanol in every washing, in a way that the last two wash is carried out without butanol.
  4. Dialyzing with a 3500 Dalton dialysis tube for 4 days with low salts physiological buffer at physiological pH.
  5. To prepare 500 ml Lipid-Depleted Fetal Horse Serum (LD-FHS), 10 changes of 10 liters of buffer is needed.
  6. The dialysis will be terminated when the buffers show no isopropanol using GC–MS.
  7. The serum  goes through a vigorous analysis for pathogens as well as growth and MTT (proliferation ability) using 2 cell lines. For complete details, please see below: quality control as well as “Analysis Tab”).
  8. The resulting high-quality serum lacks most of lipids and can still be used to support the growth and maintenance of many cell lines in culture.
