Description

Background

Since serum is usually present in cell culture studies, its lipid metabolism makes studying the role of serum in cultured cells very challenging.  That is because serum contains a wide range of undefined lipid species. PurMa™ Lipid-Depleted Fetal Horse Serum (LD-FHS) performs as the highest level of lipid depletion of serum . Lipid-Depleted Fetal Horse Serum (LD-FHS) performs crucial roles in:

  1. Where the normal concentration of lipids may interfere with the results.
  2.  It is also crucial for studies directly involving lipid metabolism. Because the composition of serum can have a profound impact on important cellular checkpoints.
  3. Moreover, LD-FHS performs in investigations about the role of lipids in their corresponding intracellular pathways.
  4. Also, it acts to generate  the media with defined lipid species.

PurMa Biologics uses a cutting-edge methodology to remove lipids from serum without eliminating its ability to support the proliferation of cells in culture.

Our proprietary method includes

  1. Applying isopropyl ether and butanol with a special ratio.
  2. PurMa Biologics has spent many years determining the needed density of ether and butanol as well as the ratio of these two solvents in this process to gain the best result.
  3. 4X washed, each for 3 hours, with isopropyl ether-butanol with decreasing amounts of butanol in every washing, in a way that the last two washes are carried out without butanol.
  4. Dialyzing with a 3500 Dalton dialysis tube for 4 days with low salts physiological buffer at physiological pH.
  5. To prepare 500 ml Lipid-Depleted Fetal Horse Serum (LD-FHS), 10 changes of 10 liters of buffer is needed.
  6. The dialysis will be terminated when the buffers show no isopropanol using GC–MS.
  7. The serum  goes through a vigorous analysis for pathogens as well as growth and MTT (proliferation ability) using 2 cell lines. For complete details, please see below: quality control as well as “Analysis Tab”).
  8. The resulting high-quality serum lacks most of lipids and can still be used to support the growth and maintenance of many cell lines in culture.

Frequently asked questions

How should I store and thaw Lipid-Depleted Fetal Horse Serum (LD-FHS)?

Store at minus 20 degrees C for long term use. Thaw overnight at 2 to 8 degrees C and then bring to room temperature with occasional gentle swirling, rather than plunging straight into a 37 degree bath. Aliquot after the first thaw, because repeated freeze and thaw cycles damage growth factors and cause cryoprecipitate.

Do I need to heat inactivate it?

Only for applications sensitive to complement, such as some immunological assays. Heat inactivation at 56 degrees C for 30 minutes also reduces some growth factors, so for routine culture most labs use serum without it. Test both if your cells are sensitive.

What testing is performed on each lot?

Each lot is tested for sterility, mycoplasma and endotoxin, along with a biochemical profile and haemoglobin. Results are reported on the Certificate of Analysis for that specific lot.

What are the flakes or particles in my thawed serum?

Almost always cryoprecipitate, which is fibrin and lipoprotein coming out of solution during freezing and thawing. It is normal, does not indicate contamination, and does not affect performance. It can be removed by filtration if it interferes with your assay.

Can I reserve a specific lot for repeat orders?

Yes. Labs that validate a lot for a sensitive application can reserve volume from it. Request lot reservation through the bulk pricing form on this page and state the volume and timescale you need.
