Background; Dulbecco’s Modified Eagle’s Medium
Dulbecco’s Modified Eagle’s Medium (DMEM) is a modification of Basal Medium Eagle (BME). Currently, DMEM is one of the most widely used media.
Major modifications compared to the original formulation:
- Although the original Dulbecco’s Modified Eagle’s Medium contained 1000 mg/L of glucose, there are different Glucose amounts used. As shown below, PurMa Biologics DMEM contains 4.5 g/L glucose. Additionally, PurMa Biologics offers a (1.5 g/L) low glucose (P3SO28105) condition.
- Furthermore, DMEM contains fourfold more amino acids and vitamins compared to the original formulation. DMEM is a good choice for growing the primary cell culture of mouse and chicken cells. Moreover, for a variety of normal and transformed cells, this media can be used properly. Each of these DMEMs, by comparison, offers a different combination of L-glutamine and sodium pyruvate.
- More importantly, DMEM has several fold more buffering capacities (3.7g/L NaHCO3) Need of FBS.
Lastly, DMEM Contains:
Furthermore, DMEM contains no proteins, lipids, or growth factors. Therefore, it requires supplementation, commonly with 10% Fetal Bovine Serum (FBS).

- Phenol red (0.0015g/L)
- L-glutamine (0.585 g/L)
