Description
PurMaTM Hybridoma Cryoprotective Media (HCM-D)
Background
PurMa™ Hybridoma Cryoprotective Media with DMSO (HCM-D) is specialized for Fused myeloma , CHO cells as well as humanized Hybridomas. It contains a proprietary combination for the best hybridoma cryopreservation, survival, and the recovery after thawing.
Why Cryopreservation
During cryopreservation cooling aqueous solutions below the equilibrium freezing point will inevitably result in the formation of ice crystals in the extracellular media. This will results in a decrease in the concentration of water which has two key consequences:
- Osmotic gradient forms across the cell membrane, which causes “cell dehydration”, and
- Ice formation and abnormal concentration of solutes in the intracellular matrix, osmatic shock, and toxicity.
DMSO benefits cryoprotection and cryoprotective agents in several ways
- Provides strong molecular stability.
- Besides, facilitates a high non-conductor constant.
- It contains the basic properties, salts solubility particularly for anions. Additionally contains a propensity to act as the acceptor atom in hydrogen bonding.
- Especially, DMSO makes a hydrogen bond with water which is 30% stronger than between two water molecules.
The proprietary formulation of PurMaTM HCM allows this media to get inside the cells instantly and lower cell death.
Special Features
- Ready-to-Use medium.
- Ready-to-Use.
- PurMaTM Hybridoma Cryoprotective Media (HCM) with DMSO provides much higher cell viability after short/long-term cryoprotection.
- PurMaTM HCM Prevents the formation of crystals in extracellular space. This, consequently, prevents cellular dehydration and resulting in less exposure to toxins from the non-recovering cells.
- Preserves morphological appearance as a standard factor of cell recovery.
- Tested for pH, osmolality, sterility, and endotoxin.
- Each Lot of PurMaTM HCM goes through rigorous testing for pathogens and sterility. We evaluate each Lot as well for several criteria. Including survival (by Trypan Blue method), viability & performance (by MTT Test), and the recovery rate by Alamar Blue test.
- PurMaTM HCM has been tested successfully for several antibody making cell lines including fused NS1 and CHO cells.
- Suitable for clinical applications.
